五爪金英Tithonia diversifolia為菊科(Compositae)植物,為台灣之民間用藥,被用於治療肝病,近年來許多報告已經證實具有保肝、抗發炎及抗腫瘤活性,本實驗室發表一篇有關五爪金英Td-F2萃取物誘導人類肝癌細胞Hep3B細胞週期阻滯及細胞凋亡作用,目前尚無五爪金英抗乳癌的相關研究。
五爪金英Td-F2萃取物,處理MCF-7乳癌細胞24小時細胞毒性試驗(MTT assay),顯示其IC50為3.589 µg/mL,Tamoxifen為22.368 µg/mL,而五爪金英Td-F2萃取物及Tamoxifen混合藥劑IC50為2.106 µg /mL;經48小時處理細胞,五爪金英Td-F2萃取物其細胞毒性IC50為2.446 µg/mL,Tamoxifen為11.873 µg/mL,而兩種混合藥劑,細胞毒性IC50為1.507 µg/mL,證明五爪金英 Td-F2萃取物對MCF-7乳癌細胞之毒殺作用,具有時間及濃度的依賴性,尤其以五爪金英Td-F2萃取物及Tamoxifen混合藥劑的結果更具有顯著的意義。
顯微鏡觀察,發現MCF-7乳癌細胞外觀型態改變。使用DNA膠體電泳、ELISA kit及自動細胞分析儀測定,結果顯示具有誘導人類乳癌細胞MCF-7凋亡現象。
綜合上述實驗結果,五爪金英Td-F2萃取物具有誘導人類乳癌細胞MCF-7凋亡作用;天然藥物-五爪金英與Tamoxifen合併使用,更具有加乘作用,對於減少乳癌患者長期使用Tamoxifen,導致子宮內膜癌的風險,在未來是可以期許的治療方向。

Tithonia diversifolia, which belongs to Compositae family, is a popular Taiwanese folk medicine, and has traditionally been used for treating liver disease. In recent years, many reports have confirmed that Tithonia diversifolia indeed shows hepatoprotective, anti-inflammatory, and anti-tumor activities. Specifically, our group has shown that Tithonia diversifolia Td-F2 extracts induce cell cycle arrest and apoptosis of Hep3B human hepatoma cells. However, until now there is no anti-breast cancer research on Tithonia diversifolia.
In our experiments, after treating MCF-7 breast cancer cells with Tithonia diversifolia Td-F2 extracts for 24 hours, cytotoxicity test (MTT assay) displayed an IC50 of 3.589 μg/mL, as compared with 22.368 μg/mL and 2.106 μg/mL for those treated with Tamoxifen and the confection of Tithonia diversifolia Td-F2 extract + Tamoxifen , respectively. In addition, by prolonging the treatment time to 48 hours, cytotoxicity test (MTT assay) displayed an IC50 of 2.446 μg/mL, 11.873 μg/mL, and1.507 μg/mL for Tithonia diversifolia Td-F2 extract, Tamoxifen, and the confection of Tithonia diversifolia Td-F2 extract + Tamoxifen, respectively. These results indicate that the toxicity of Tithonia diversifolia Td-F2 extracts shows time and concentration-dependency. The pharmaceutical effectiveness is particularly significant for the confection of Tithonia diversifolia Td-F2 extract + Tamoxifen, among the three splits studied.
In addition, we also performed microscopic observations and confirmed the morphological changes for the appearance of the Tithonia diversifolia Td-F2 extracts-treated MCF-7 breast cancer cells. Finally, all results from DNA gel electrophoresis, ELISA kit, and automatic cell analyzer confirmed that Tithonia diversifolia Td-F2 extracts indeed induce cell apoptosis.
In summary, Tithonia diversifolia Td-F2 extracts show significant toxic effects and induce apoptosis in MCF-7 human breast cancer cells. The toxic effects are especially significant synergistic by confection Tithonia diversifolia Td-F2 extracts with Tamoxifen. The confection therefore appears to be very promising for reducing the risk of uterine cancer in conventional adjuvant therapy using pure Tamoxifen, and is a potential cancer chemo-preventive agent.
