中 文 摘 要
在工業化國家中,心血管疾病(包括高血壓、動脈粥樣硬化、心肌梗塞和腦中風等)的死亡率極高,使得心血管疾病的成因和影響因素在最近二十年來被大力的研究。1970年代的研究即已發現,葡萄酒攝取與心血管疾病死亡率有顯著負相關的關係存在。 近年來的研究又發現,紅葡萄酒可使大白鼠的血管平滑肌產生舒張作用,紅葡萄酒中的重要成分檞皮酮在血管平滑肌產生舒張作用時扮演著重要角色。因此本實驗在探討檞皮酮對於大白鼠,血管平滑肌的作用機轉。離體實驗部份,將取自正常血壓鼠或L-NAME所誘導之高血壓大白鼠的胸主動脈,連接於張力轉能器,記錄對藥物的收縮或舒張反應。結果顯示,在無內皮細胞存在的狀況下,檞皮酮(10-4 M)能抑制血清胺所引起的血管收縮反應,但檞皮酮(10-4 M)卻不能抑制phenylephrine所引起的血管收縮反應。由本論文的結果顯示,檞皮酮選擇性抑制血清胺收縮反應的機轉,無法被一氧化氮合成酵素抑制劑或前列腺素合成酵素抑制劑所阻斷。所以檞皮酮可能不是經由一氧化氮或是前列腺素來達成抑制血清胺所引發的收縮反應。
慢性腹腔給予檞皮酮(四週)雖無法抑制L-NAME所誘發的高血壓,但可使高血壓鼠的血管對phenylephrine 的收縮性顯著被抑制。檞皮酮由於可抑制血清胺(急性效果)及phenylephrine(慢性效果)的收縮反應,因此檞皮酮在治療心血管疾病的療效上值得進一步去探討。

Abstract
Since the early 1970s, there have been increasing evidences that the consumption of moderate amounts of red wine is inversely correlated with the mortality of myocardial infarction. Quercetin, the major component of the red wine may be involved in the prevention of cardiovascular diseases. In an isolated vessel preparation, aortic rings from Sprague-Dawley rats or L-NAME induced hypertensive rats were placed in tissue baths to record isometric tension. Our results demonstrated that quercetin (10-4 M) selectively inhibits the vascular contraction induced by serotonin (10-10 ~3 X 10-5 M). In contrast, quercetin did not inhibit the vascular contraction induced by phenylephrine. This inhibitory effect of quercetin on serotonin-induced vascular contraction was not prevented by pretreatment the vessels with indomethacin or L-NAME. Our results indicate that nitric oxide and prostaglandin may not involve in the acute effects of quercetin on serotonin-induced vascular contraction. In chronic L-NAME induced hypertensive rats, daily injection of quercetin did not prevent the progress of hypertension. However,chronic quercetin injection inhibited the vascular contraction induced by phenylephrine in L-NAME induced hypertensive rats. In summary, quercetin acutely inhibited serotonin-induced vascular contraction and chronically inhibited phenylephrine-induced vascular contraction suggesting a potential role for further clinical application.
