中醫藥學在近年學理驗證的努力下,已逐漸受到全世界的重視與肯定,目前正是中醫藥學蓬勃發展的時代。許多文獻記載,多數的中藥都有降低發炎反應功效。中藥複方劑黃耆建中湯 (Huang-Qi-Jian-Zhong-Tong; HQJZT) 是由黃耆、桂枝、甘草、大棗、芍藥、生薑、膠飴等藥材所組成,很多研究顯示這些藥材都具有抗發炎及增強免疫力之功能。本研究探討中藥材黃耆建中湯原料、藥渣、青貯藥渣是否可抑制細胞發炎作用,藥渣的部分是否能作為動物餵飼以及蒸煮過後的藥渣是否會影響成分。本實驗先利用黃耆建中湯藥渣餵食公羊9週,於定期抽取血液,分析包括尿素氮、血糖、白蛋白、血紅素、血小板等等其生化檢驗,探討藥渣對羊隻之生理影響,結果顯示黃耆建中湯藥渣不會增加羊隻生理負擔,綜合以上許多檢驗結果得知,黃耆建中湯藥渣不影響羊隻生理活性,可應用於動物的餵飼。再來針對黃耆湯建中湯藥渣是否有殘留的功效做分析,以HPLC分析黃耆建中湯藥渣、原料、青貯藥渣中毛蕊異黃酮含量,但結果顯示毛蕊異黃酮在黃耆建中湯原料中含量較高但在藥渣與青貯中含量卻下降,青貯蒸煮後會降低毛蕊異黃酮的含量。為了更快速分析黃耆建中湯藥渣是否具有降低發炎反應,利用黃耆建中湯原料、藥渣、青貯藥渣之萃取物處理小鼠巨噬細胞株 (macrophage cell line RAW264.7)分析其存活率及分泌一氧化氮的影響,將脂多醣(Lipopolysaccharide, LPS) 處理RAW264.7,使細胞受脂多醣刺激而分泌大量一氧化氮,再利用加入不同濃度的黃耆建中湯原料、藥渣、青貯藥渣之萃取物,顯示其可抑制發炎現象且不會造成細胞死亡。黃耆建中湯抑制發炎的作用機制仍需更多細胞內蛋白質的分析。
In recent years, Chinese medicine theoretical verification efforts, has gradually attention and recognition around the world, now is the era of vigorous development of Chinese medicine. Many documented, most Chinese have to reduce inflammation effect. Chinese medicine Huang-Qi-Jian-Zhong-Tong(HQJZT); HQJZT is composed of Astragalus, cassia twig, licorice, jujube, peony, ginger, gum syrup and other ingredients consisting of a lot of studies have shown that these medicines They have anti-inflammatory and enhance the immune system's function. In this study, HQJZT dregs, silage dregs whether inhibit cell-inflammatory effects, whether or not the dregs section can be used as animal feed and residue after cooking will affect the composition. In this study, the first use of HQJZT dregs feeding ram nine weeks, at regular sampling of blood, analysis includes urea nitrogen, glucose, albumin, hemoglobin, platelets, and so its biochemistry, physiology sheep explore the impact on the dregs, the results showed that HQJZT dregs which can lower blood sugar, kidney function does not affect the physiological burden increase sheep, integrated over a number of test results that, HQJZT dregs of sheep does not affect the physiological activity, it can be used in animal feed . Whether coming for HQJZT dregs have residual effect for analysis by HPLC analysis HQJZT dregs, feed, silage dregs calycosin content, but the results show calycosin soup ingredients in Astragalus Jianzhong However, with higher levels in the content of the dregs and fell silage will reduce the content of silage calycosin cooking. In order to more quickly analyze whether HQJZT dregs reduce inflammation, use extraction Huang-Qi-Jian-Zhong-Tong dregs, silage dregs of the treated mouse macrophage cell line (macrophage cell line RAW264.7) to analyze the survival rate and secretion of nitric oxide, the LPS (Lipopolysaccharide, LPS) treated RAW264.7, the cells were stimulated by LPS secrete large amounts of nitric oxide, and then use different concentrations of HQJZT, dregs, silage dregs, the show that inhibits inflammation and will not cause cell death. Analysis of intracellular proteins Astragalus Daikenchutou suppress inflammation mechanism still needs more.
