金銀花具有清熱解毒之功效且含有豐富多酚類成分。經益生菌發酵可能具有提高藥效或生物利用度等效益。乳酸菌與酵母菌為常見的益生菌,並於共培養時可提升產物的含量。疲勞會造成反應能力、工作效率的下降。長時間的疲勞亦會造成體內器官的衰退。補充多酚可提升肌肉抗氧化能力、增加游離脂肪酸含量達到延遲疲勞及抗疲勞之效果。本研究探討乳酸菌及酵母菌共培養金銀花葉萃取物發酵過程中多酚類的成分變化,並利用反應曲面法探討乳酸菌與酵母菌對金銀花葉萃取物發酵的條件,最後再經由動物實驗評估發酵物之抗疲勞功效。經由高效能液相層析儀 (high performance liquid chromatography, HPLC) 分析結果可發現經由益生菌發酵後,其兒茶酚、兒茶素與綠原酸皆有上升。Lactobacillus plantarum 與 Saccharomyces cerevisiae 共培養可增加益生菌的存活。而共培養系統發酵金銀花葉萃取物在 12 hr、24 hr 時所含兒茶酚及兒茶素含量較高,其綠原酸含量於 24 hr、30 hr 亦較高,因此選擇以共培養方式進行實驗。經由第二部份結果得知於起始 pH 6.5、 培養時間 21 hr、培養溫度 23oC 為發酵的最適條件。經由驗證在起始 pH 6.5、培養溫度 23oC、培養時間 21 hr 條件下可得兒茶酚 127 μg/mL、兒茶素 228 μg/mL 與綠原酸 176 μg/mL。相較於未發酵之金銀花葉萃取物,兒茶酚及兒茶素有顯著的提高 (p < 0.05)。第三部分抗疲勞功效評估,發現餵食金銀花葉益生菌發酵物可使其游泳時間增加、提升肌力回復、促進游離脂肪酸提升、提升乳酸的代謝率、降低平時的血氨濃度並提升肌肉肝醣含量以及肝臟中 monocarboxylate transporter 4 (MCT 4) 蛋白表現。綜合上述結果,經由L. plantarum 與 S. cerevisiae 對金銀花葉萃取物於起始 pH 6.5、培養時間 21 hr、培養溫度 23oC 可獲得最大產量而兒茶酚由 49.2 μg/mL 提升至 127 μg/mL、兒茶素由 105 μg/mL 提升至 228 μg/mL、綠原酸由 175 μg/mL 提升至 176 μg/mL。餵食發酵物則可藉由提升肌肉肝醣,增加肌力回復。並藉由提升肝臟中 MCT 4 蛋白表現而提升乳酸的代謝率,並促進游離脂肪酸提升、降低平時的血氨濃度,進而提升基礎體力及疲勞回復,其主要功效來自於兒茶素。

Honeysuckle containing rich chlorogenic acid and other polyphenol constituents was proven to provide anti-inflammatory and antiviral effect. It may have higher bioavailability via probiotic fermentation. Lactobacillus and yeast are familiar probiotics and their products content can be increased by their co-culture. Fatigue reduces response capability and working efficiency. Long-term fatigue leads to organs recession. However polyphenol supplementation was found to increase muscle antioxidant and free fatty acid content and further perform anti-fatigue. In this study, the change of polyphenol during the honeysuckle leaf extract fermentation by Lactobacillus and yeast was investigated, and the optimum fermentation condition was studied by Response surface methodology (RSM). The anti-fatigue effect was for the investigated by animal test. The results analyze by high performance liquid chromatography (HPLC) indicated that catechol, catechin, and chlorogenic acid content were increased. The viability of catechol and catechin content was improved. The fermented honeysuckle leaf extract by co-culture included high content of catechol and catechin at 12 hr and 24 hr as well as high content of chlorogenic acid at 24 hr and 30hr. Therefore co-culture was selected in this study. In the second phase the optimum fermentation condition initial pH 6.5, 23oC, 21 hr of culture time, was chosen.
